SHOOT INDUCTION OF STRAWBERRY cv. DORIT THROUGH TDZ PRE-TREATMENT AND STEM TRANSVERSE SECTION METHOD

The demand of high-quality seedling on strawberry prodution in large quantity is a necessity to optimise maximal gain in the field production. In vitro meristem culture of Strawberry has successfully been established, however a reliable approach to speed up the shoot multiplication rate needs further investigation. The aim of present study was to investigate the effect of various concentrations and time exposures of pulsing TDZ solution towards stem nodus explant in combination with the transverse section technique on adventitious shoot multiplication of Strawberry cv. Dorit. The experiment was conducted at Indonesian Citrus and Subtropical Fruits Research Institute from June to August 2021. The result showed that pulsing the nodus explant without transverse section (TS) in 1 mg/l TDZ solution for 1 hour yielded higher number of adventitious shoots at 9.4 shoots per explant with the absent of visible callus formation compared to 5 hour exposure to TDZ. Transverse section (TS) is evidently increasing the adventitious shoot per cm explant length of with the total value of 14.8 shoots.
